ABSTRACT

Os diferentes grupos de E. coli podem estar presentes no leite e seus derivados, que podem resultar em infecções em humanos. Este estudo objetivou isolar E. coli de diferentes pontos da obtenção do leite e da elaboração de queijos tipo Minas frescal, detectar os patótipos EAEC, EIEC, ETEC, EPEC, STEC, ExPEC e caracterizar os isolados pela pesquisa de genes de virulência . Para tanto, foram realizadas coletas em cinco pequenas propriedades rurais produtoras deste tipo de queijo no nordeste do estado de São Paulo. Foram coletadas amostras de suabes de fezes bovinas, de mãos de ordenhador, balde, leite, soro, água, superfície de elaboração de queijos, mãos de manipulador do queijo, peneiras, bandejas, fôrmas e escumadeiras. Foram obtidos 73 isolados de E. coli potencialmente patogênicos, sendo que nas amostras de leite e queijo foram encontrados isolados como STEC e ExPEC. Assim, a presença de cepas de E. coli potencialmente patogênicas na obtenção do leite e na produção do queijo constitui risco para a saúde pública.

ABSTRACT

El pangenoma de Escherichia coli se compone de un core conservado y regiones genómicas variables. El componente genético constante permite determinar la filogenia del microorganismo, mientras que la variabilidad genética ha permitido el surgimiento de cepas patógenas intestinales y extraintestinales. En este estudio caracterizamos genéticamente 85 cepas patógenas extraintestinales aisladas de caninos y felinos. Se utilizó el esquema de Clermont para agruparlas en relación con elfilogrupo de pertenencia (intestinal: Ay B1; extraintestinal: B2 y D) y se investigó en ellas la presencia de varios marcadores de virulencia (pap1-2, pap3-4, sfa, afa, hlyA, aer y cnf), así como de marcadores de patovares híbridos. El 69,4% de los aislamientos pertenecieron al filogrupo A; el 1,2% al B1; el 16,5% al B2 y el 12,9% al D. El gen encontrado con mayor frecuencia fue sfa (21/85), seguido de los genes pap1-2 y cnf (20/85) y pap3-4 (19/85). No se detectaron híbridos. Los aislamientos en animales deben ser estudiados debido al potencial zoonótico del microorganismo.


The pangenome of Escherichia coli is composed of a conserved core and variable genomic regions. The constant genetic component allows to determine the phylogeny of the microorganism, while genetic variability promoted the emergence of intestinal pathogenic strains and extraintestinal strains. In this study we characterized 85 extraintestinal pathogenic isolates genetically isolated from canines and felines. We used the Clermont scheme that includes intestinal (A and B1) and extraintestinal (B2 and D) phylogroups, virulence markers (pap1-2, pap3-4, sfa, afa, hlyA, aer and cnf) and hybrid pathogens. A percentage of 69.4% of the isolates belonged to phylogroupA; 1.2% to phylogroup B1; 16.5% to phylogroup B2 and 12.9% to phylogroup D. The most commonly found gene was sfa (21/85), followed by pap1-2 and cnf (20/85) and pap3-4 (19/85). No hybrids were detected. Animal isolates should be studied due to the zoonotic potential of the microorganism.

ABSTRACT

In this study, avian extraintestinal Escherichia coli obtained from the liver of poultry carcasses approved for human consumption in the State of Pernambuco-Brazil were tested for antibiotic plus serum-resistance. Liver samples (n=110) were obtained from one slaughterhouse and 88 bacterial isolates were identified as Escherichia coli. The antibiotic-resistance profiles of antibiotics used in human and/or veterinary practice were accessed by the disk-diffusion method. Phenotypes with high resistance to streptomycin (84.0%), tetracycline (44.7%), amikacin (29.8%), gentamicin (21.3%) and ciprofloxacin (21.3%) were identified. Resistance to antibiotics such as ceftazidime, amoxicillin-clavulanic acid and imipenem was also recorded. Twenty isolates with distinct antibiotic-resistance and susceptibility profiles were selected for serum resistance assays, phylogenetic characterization and detection of the iss gene. We have shown that multidrug resistant isolates were often simultaneously resistant to broiler and human sera. Phylogenetic characterization of serum- plus antibiotic-resistant isolates have shown three belonging to group D, eleven to group B1, one to group B2, and five to group A. We concluded that commensal E. coli strains isolated from the liver of healthy poultry carcasses can harbor and potentially share multidrug- plus virulence genes found in pathogenic pathotypes. This suspicion was not related to specific phylogenetic groups or presence of the iss gene.(AU)


Neste estudo, isolados de Escherichia coli extraintestinal aviária obtidos a partir do fígado de carcaças de aves aprovadas para consumo humano no Estado de Pernambuco-Brasil foram testados para resistência a antibióticos e soro. As amostras de fígado (n = 110) foram obtidas de um abatedouro, sendo 88 isolados bacterianos identificados como Escherichia coli. Os perfis de resistência a antibióticos de uso humano e/ou veterinário foram determinados pelo método de disco-difusão. Foram identificados fenótipos com alta resistência à estreptomicina (84,0%), tetraciclina (44,7%), amicacina (29,8%), gentamicina (21,3%) e ciprofloxacina (21,3%). A resistência a antibióticos utilizados na medicina humana e/ou veterinária, tais como a ceftazidima, amoxicilina-ácido clavulânico, estreptomicina e imipenem também foi registrada. Vinte amostras com perfis distintos de resistência/sensibilidade a antibióticos foram selecionadas para os ensaios de resistência ao soro, caracterização filogenética e detecção do gene iss. Foi demonstrado que isolados resistentes a múltiplas drogas foram também simultaneamente resistentes ao soro de frangos e ao soro humano. A caracterização filogenética desses isolados mostraram três pertencentes ao grupo D, onze ao grupo B1, um ao grupo B2 e cinco ao grupo A. Conclui-se que E. coli comensais isoladas do fígado de carcaças de aves saudáveis podem abrigar e potencialmente compartilhar genes de resistência a drogas e de virulência encontrados em patotipos patogênicos. Essa suspeita não foi relacionada com grupos filogenéticos específicos ou com a presença do gene iss.(AU)

Phylogenetic analysis has shown that ExPEC is composedof four main phylogenetic groups (A,B1, B2, and D) and that virulent extraintestinalstrains mainly belong to groups B2 and D.In this study, we aimed to assess therelation between adherence virulence and phylogenetic groups of ExPEC.A total of 161 E.coli samples were collected. Out of these 17 (10.6%) werefrom pus, 66 (41 %) from urine, 78 (48.4%) from cervical swab. The phylogeneticgroups and 6 virulence genes (fimH, papC, papGII, papGIII, fa/draBC,andSfa/focDE) encoding adhesins were identified by triplex PCR. Phylogeneticgroups distribution was as follows: B1 10.5%, A 24.7%, B2 25.3%, and D 38.9%. Virulence genes prevalence was fimH 90.1%, papC 23%, papGII 16.8%, papGIII1.9%, Afa/draBC 11.8%, andSfa/focDE 5.6%. The cell surface protein (curli) wasdetected 50,3% by Congo red agar. In conclusion: The most isolated strainsbelonged to the phylogenetic group B2 and D. The phylogenetic groups weresignificantly associated with some genes encoding adhesins (fimH, papC) and cellsurface protein (curli).

ABSTRACT

Determinadas linhagens de Escherichia coli comportam-se como patógenos em gatos, causando doenças gastrointestinais e extraintestinais. Neste estudo, foram utilizadas 205 cepas de E. coli isoladas de amostras de fezes provenientes de 19 gatos diarreicos e de 21 gatos não diarreicos, e três amostras de urina provenientes de gatos com infecção do trato urinário (ITU). Essas cepas foram testadas pela técnica de reação em cadeia da polimerase com múltiplos iniciadores para a detecção da presença de genes codificadores de adesinas (pap, sfa e afa), assim como para a detecção dos genes produtores da toxina Shiga-like (stx1 e stx2) e do gene da intimina (eae). Oitenta e dois isolados possuíam genes codificadores de adesinas, dos quais 11 apresentaram o gene pap, 41 apresentaram o gene sfa e 27 apresentaram uma combinação dos genes pap + sfa. Nenhuma das cepas examinadas apresentou os genes stx1, stx2 ou afa. Três isolados provenientes de um gato diarreico apresentaram uma combinação dos genes sfa + eae. Animais de companhia (pets) são reservatórios naturais para diversos organismos, especialmente linhagens ExPEC, as quais são potencialmente capazes de infectar seres humanos, o que representa um motivo de grande preocupação.(AU)

ABSTRACT

The soft cheese Minas frescal is one of the most popular cheese in Brazil, which is typically manufactured in small dairy farms under unsatisfactory hygiene conditions. To assess the risk involved in consumption of this cheese, virulence markers were investigated in 330 Escherichia coli strains isolated from 30 Minas frescal cheeses inspected by official government agency (SIF - serviço de inspeção federal), from 50 cheeses not inspected by SIF and 31 cheeses not inspected by SIF with spice added, all of them collected in the southwest of Minas Gerais State. The E. coli isolates were screened for the presence of Shiga toxin-encoding (stx 1 and stx 2), intimin (eae) genes and for the presence of (pap, sfa, afa) genes related to adhesion in epithelial cells. The only gene detected by PCR was the sfa gene at one isolate. The strains were also screened for resistance to 9 antimicrobial drugs. Predominant resistance was to cephalothin, tetracycline and streptomycin. Multidrug resistance was found among isolates from cheese with SIF (16.6%), cheese without SIF (8.0%) and cheese without SIF with spice added (30.0%) what is a reason for concern due to the high consumption of raw milk cheese by the Brazilian population.


O queijo Minas frescal é um dos queijos mais populares no Brasil, ele é tipicamente manufaturado em pequenas fazendas de leite, em condições higiênicas insatisfatórias. Para verificar o risco envolvido no consumo deste queijo, marcadores de virulência foram investigados em 330 cepas de isoladas de 30 queijos Minas frescal, inspecionados pela agencia governamental oficial (SIF- serviço de inspeção federal), de 50 queijos não inspecionados pelo SIF e de 31 queijos não inspecionados pelo SIF e com condimentos adicionados. Todos eles foram coletados na região sudeste do Estado de Minas Gerais. As cepas de E. coli foram examinadas para a presença de genes codificadores de Shiga toxina (stx1 e stx 2), gene da intimina (eae) e também para a presença dos genes (pap, sfa, afa) relacionados com a adesão em células epiteliais. O único gene detectado por PCR foi sfa em um isolado. As cepas também foram examinadas para a resistência a nove fármacos antimicrobianos. As resistências predominantemente detectadas foram para cefalotina, tetraciclina e estreptomicina. A resistência a múltiplas drogas identificadas entre as cepas de E. coli, provenientes de queijo com SIF (16,6%), queijo sem SIF (8,0%) e queijo sem SIF com condimentos adicionados (30,0%), o que representa um motivo de preocupação, devido ao elevado consumo pela população Brasileira de queijos feitos de leite não pasterurizado.

ABSTRACT

The zoonotic potential to cause human and/or animal infections among multidrug-resistant extraintestinal pathogenic Escherichia coli from avian origin was investigated. Twenty-seven extraintestinal pathogenic E. coli isolates containing the increased survival gene (iss) were obtained from the livers of healthy and diseased poultry carcasses at two slaughterhouses in Salvador, northeastern Brazil. The antimicrobial resistance-susceptibility profiles were conducted with antibiotics of avian and/or human use by the standardized disc-diffusion method. Antimicrobial resistance was higher for levofloxacin (51.8%), amoxicillin/clavulanic acid (70.4%), ampicillin (81.5%), cefalotin (88.8%), tetracycline (100%) and streptomycin (100%). The minimum inhibitory concentrations above the resistance breakpoints of doxycycline, neomycin, oxytetracycline and enrofloxacin reached, respectively, 88.0%, 100%, 75% and 91.7% of the isolates. Strains with high and low antimicrobial resistance were i.p. administered to Swiss mice, and histopathological examination was carried out seven days after infection. Resistance to goat and human serum complement was also evaluated. The results show that Swiss mice challenged with strain 2B (resistant to 11 antimicrobials) provoked a severe degeneration of hepatocytes besides lymphocytic infiltration in the liver, whereas the spleen showed areas of degeneration of the white and red pulp. Conversely, the spleen and liver of mice challenged with strain 4A (resistant to two antimicrobials) were morphologically preserved. In addition, complement resistance to goat and human serum was high for strain 2B and low for strain 4A. Our data show that multidrug resistance and pathogenesis can be correlated in extraintestinal pathogenic E. coli strains obtained from apparently healthy poultry carcasses, increasing the risk for human public healthy.

ABSTRACT

From January to December 2006, 92 Escherichia coli isolates from 25 diarrheic dogs were analyzed by screening for the presence of adhesin-encoding genes (pap, sfa, afa), hemolysin and aerobactin genes. Virulence gene frequencies detected in those isolates were: 12 percent pap, 1 percent sfa, 10 percent hemolysin and 6.5 percent aerobactin. Ten isolates were characterized as extraintestinal pathogenic E. coli (ExPEC) strains; all showed a multidrug resistance phenotype that may represent a reason for concern due the risk of dissemination of antimicrobial resistant genes to the microbiota of human beings.


Entre Janeiro e dezembro de 2006, 92 cepas de Escherichia coli isoladas de 25 cachorros diarréicos foram analisadas para a detecção de genes codificadores de adesinas (pap, sfa e afa), hemoisina e aerobactina. As freqüências dos genes de virulência detectadas nestas cepas foram: 12 por cento pap, 1 por cento sfa, 10 por cento hemolisina e 6,5 por cento aerobactina. Dez cepas foram caracterizadas como E. coli extraintestinal patogênica (ExPEC) e todas as cepas apresentaram um fenótipo de multiresistência, o que pode representar um motivo de preocupação, por causa do risco de disseminação de genes de resistência a drogas antimicrobianas para a microbiota dos seres humanos.

ABSTRACT

The study was conducted in twenty-three butcheries in the city of Taquaritinga, State of São Paulo, Brazil, surveyed during a 10 months period. Among two hundred and eighty-seven Escherichia coli strains isolated from samples of ground beef, meat-grinding-machines and the hands of manipulators, five were recognized as extraintestinal pathogenic E. coli (ExPEC), showing virulence factors (P and S fimbriae, hemolysin and aerobactin) and presenting multidrug resistance. Retail-sold food may constitute an important vehicle for the dissemination of ExPEC in communities, giving rise to reasons for concern.


O trabalho foi desenvolvido em 23 açougues em Taquaritinga, Estado de São Paulo, Brasil, durante um período de 10 meses. De duzentas e oitenta e sete cepas de E.coli isoladas de carne moída, moedor de carne e mãos de manipuladores de carne, cinco foram caracterizadas como E. coli patogênica extraintestinal (ExPEC) apresentando fatores de virulência (fimbria P e S, hemolisina e aerobactina), assim como multiresistencia a drogas antimicrobianas. Retalhos de carne podem ser um veiculo importante para a disseminação de ExPEC, o que representa um motivo de preocupação.
